A double beam UVVis spectrophotometer is an advanced analytical instrument used to measure the absorbance or transmittance of substances in the ultraviolet 200400 nm and visible 400800 nm regions of the electromagnetic spectrum It is widely used in chemistry biochemistry pharmaceuticals and environmental analysis for quantitative and qualitative analysis of compounds

In a double beam system the light from a source such as a deuterium lamp for UV and a tungsten lamp for visible light is passed through a monochromator which selects a specific wavelength The monochromatic light is then split into two beams using a beam splitter

One beam passes through the sample solution

The other beam passes through a reference or blank solution

Both beams are directed to detectors and their intensities are measured simultaneously or alternately The instrument compares the intensity of light passing through the sample I with that of the reference I and calculates absorbance using BeerLamberts law
